Description: Currently, the LPDNA has three new generation sequencers acquired by the Federal University of Pará: SOLiD version 3 plus and SOLiD 5500, these technologies being able to generate data with accuracy of up to 99.99%, a throughput of 60 - 90 Gb per run, generating bases with Phred value of quality 30 in version 3 plus and 40 or more for version 5500; the third device is the Ion Torrent, a recent platform launched on the market, which has some advantages over other new generation technologies such as reduced running time and low cost. The availability of these three devices allows us to sequence complete genomes from fragment, mate-paired and paired-end libraries. Among the applications that can be performed on the LPDNA through the SOLiD platform are: i) Genomics; ii) Sequencing again; iii) Targeted Resequencing; iv) Whole Genome Resequencing; v) Transcriptomics; vi) Gene Expression Profiling (SOLiD - SAGE [Serial Analysis of Gene Expression] System) for the quantification of gene expression; vii) Small RNA Analysis; viii) Whole Transcriptome Analysis; ix) Epigenome; x) Chromatin Immunoprecipitation Sequencing (ChIP-Seq); xi) Methylation Analysis. The LPDNA also has infrastructure for the large-scale analysis of proteins from microorganisms of great biotechnological value. The laboratory has high sensitivity and precision equipment that allows a complete analysis of gene expression using DIGE technology in conjunction with MALDI-TOF mass spectrometry. Through this approach, it is possible to analyze the molecular behavior of microorganisms exposed to different chemical, biochemical and physical stimuli, identifying and characterizing peptides of biotechnological interest.

Bioinformatics Unit: Area of ​​approximately 50m2. The computational infrastructure that supports the bioinformatics activity at UFPA consists of a server with two INTEL QUAD-CORE XEON E5440 2.83 GHz processors, 24 Gb of RAM and 1 Tb of hard disk, a storage unit with 7 Tb , a test server with a Core2Duo processor, 500 GB of hard disk and 4 GB of RAM, in addition to 11 workstations, each with a Core 2 Duo processor and 2 GB of RAM. Regarding data integrity, management responsible for triggering backup disks in case of failure has been implemented in the storage unit, so that the data is always available. In addition, there is an exclusive area for backup outside the storage unit. Access to the server can be performed remotely, allowing data to be transferred, processed and analyzed remotely. The data of organisms with sequenced genomes are stored in a relational database, modeled to properly receive SOLID data, thus allowing quick access to information, in addition to their integration. To manage this data, the MySQL database management system is used. It is a robust database and ideal for the management of large volumes of data, such as those generated by the sequencer and subsequent analyzes.

Proteomics Unit: Physical area of ​​approximately 30m2. Bidimensional Electrophoresis Unit Ettan DALTsix (Amersham Biosciences); Two-dimensional Electrophoresis Unit Ettan IPGphor II (Amersham Biosciences); Two-dimensional image scanner for ImageScanner gel (Amersham Biosciences); Ettan 2-D Fluorescence Difference Gel Electrophoresis System - DIGE (GE Healthcare); Ettan Spotter (GE Healthcare); Ettan Digester (GE Healthcare); Ettan Spot Picker (GE Healthcare); Image scanner Ettan DIGE Imager (GE Healthcare); Software for the analysis of gels Image Master 2D Platinum v.7.0 (GE Healthcare); MALDI-TOF AXIMA-CFR plus mass spectrometer (Shimadzu).
